“two”には、「2つ、2人」という意味のほかに、
「2つ1組のもの、2人1組のもの」という意味があります。
ですから、“That makes two of us.”を直訳すると、
「それは、私たちを1組のものにする」という意味になり、そこから、
「それは私たち2人に言えることだ」「私も同じだ」「同感だ」「私も」
などという意味になります。
自分も相手と立場や意見が同じだという場合に用いられる表現ですね。
